How to tell the prescription of a contact lens

This might sound like a daft question, but can you tell what prescription a contact lens is from the lens itself? Or do you have to have the packet? I've had a lady call up to say that she thinks she's mixed her lenses up, and I don't know how to help her

I'm guessing you are referring to soft CLs, since you said packet. If you don't have a wet cell to verify...and most don't, you can carefully blot the lens dry and place it on the aperture of your lensometer to check power. Place the concave side of the lens on the aperture. This won't give an "exact" power, but you would be able to come close and by comparing the two lenses, you would be able to tell strong vs. weak. Be sure to clean the aperture well first and then clean and disinfect the lenses after.

In most cases, reversing the lenses will provide the answer- the eye that needs the most minus will be slightly blurred at infinity if reversed.

Do an over refraction. If you get plano, or close to it then you are good, if you get one eye + and the other - then they are switched (assuming you were not doing monovision)
